These are options on the backend. If these options apply depends on the setup of the theme you are using as some override the backend options for design reasons.

HTML/CSS does not know "columns". How many images you get per row is define by how many image fit into a row and that is also defined by the HTML/CSS setup of your theme. The number of images per page is also an option.

Please get a little familiar with Zenphoto in general first before trying to make a theme. Also please read our theming tutorial. HTML/CSS knowledge is of course required.
