I am using Blurb as a book printing service and only learned now that they actually offer an API, that could draw images from my Zenphoto website and then turn it into a book.

What I would need is the following: Let people mark images in Zenphoto for subsequent printing. One completed, all the images should be in some sort of basket/private album for sorting.

Then I would run a script (php) to have the book printed. Is it possible to do what is described in the previous paragraph and/or does something like this already exist?

No, we don't have such an tool. That would have needed to be coded. You could use dynamic albums as a quick workaround by using specific tags to create it. But nevertheless you will have to do some coding to get that printing service to use that correctly.
